The information from these studies provides safety
data on the persistence of the DNA in the target, such as dissemination
throughout the tissues and interaction with host DNA. The preclinical
safety evaluation of gene therapy vectors and vaccines, whether
viral, naked or complexed DNA or cell based, may rely in part on
in vivo studies in a relevant animal model. These studies are designed
to investigate a number of properties of the product and the vector
including:
- Tissue tropism and distribution by Quantitative PCR (QPCR)
- Transgene expression in tissues by quantitative RT-PCR or western blotting
- Effects on target gene expression e.g. induction of cytokines or other cellular genes
- Immunogenicity or toxicity of vector or product
